Waar ik me zoal mee bezig houd
29 maart
- freeze immature tomato tissue in liquid nitrogen
- grind the tissue with mortar (filled with liquid nitrogen) and pestle
- put the samples on ice, open the caps and add 1 mL Trizol reagent (per 50-100 mg tissue)
- close the caps, mix and incubate at room temperature for 5 minutes
- add 200 µL chloroform
- mix 15 seconds and leave at room temperature for 2-3 minutes
- spin 15 minutes at 11,000 rpm and 4 °C
- transfer the aqueous phase to a new tube
- add 500 µL isopropanol, mix well by inverting and leave at room temperature for 10 minutes
- spin 10 minutes at 11,000 rpm and 4 °C
- pour off the supernatant
- wash twice with 150 µL 75% DEPC ethanol
- spin 5 seconds at 13,000 rpm, 4 °C and remove the remaining supernatant with a yellow tip
- dry the pellets 2 minutes in the vacuum excitator
- dissolve in 40 µL DEPC water

:-O Het is wél een tabel! Wat je ook had kunnen doen was er een ‘ul’ met ‘li’-tjes tegenaan gooien, dat is simpeler. (Gewoon een tip, ik kan hier inhoudelijk toch te weinig op ingaan om er iets zinnigs over te zeggen (A) )
Eh, ohja. Dat probeerde ik eerst de editor automatisch te laten doen, maar dat mislukte, dus toen heb ik maar een tabel gemaakt. (Gedachtekronkels zijn soms raar. Dan slaan ze gewoon een stap over.)
Het klinkt alsof jij een uiterst interessant leven hebt. Ik ben dan ook zeer jaloers op jouw activiteiten.
Hihihi, ik ken veel mensen die er absoluut niet jaloers op zijn. Maar eigenlijk vind ik het zelf wel interessant. En het ‘liquid nitrogen’-gedeelte was zeker een gave activiteit (al kreeg je er wel bevroren vingers van).
